目的 利用梯度洗脱,建立同时测定复方虎黄喷剂中3种指标成分虎杖苷、黄芩苷和欧前胡素的UPLC检测方法。方法 采用UPLC法,Acquity UPLC BEH C18色谱柱(100 mm×2.1 mm,1.7 μm),流动相:乙腈(A)和0.05%磷酸水溶液(B)梯度洗脱;流速:0.4 mL/min;检测波长:280 nm;柱温:35 ℃。结果 虎杖苷的线性范围为0.59~59.0 mg/L (r=0.999 2),平均回收率为100.6%,RSD为1.56%;黄芩苷的线性范围为1.128~112.8 mg/L (r=0.999 9),平均回收率为101.5%,RSD为1.68%;欧前胡素的线性范围为0.109~10.9 mg/L (r=0.999 9),平均回收率为100.6%,RSD为2.75%。结论 本方法操作简便,测定结果准确可靠,可用于复方虎黄喷剂的质量控制。 |

OBJECTIVE To develop a quantitative method simultaneously to determinate polydatin and baicalin in compound Hu-huang Spray by UPLC. METHODS The seperation was performed on a column Acquity UPLC BEH C18(100 mm×2.1 mm,1.7 μm). The mobile phase was composed of acetonitrile (A) and 0.05% H3PO4(B) with gradient elution. The flow rate was 0.4 mL /min with the wave length at 280 nm. The temperature was 35℃. RESULTS For polydatin, the calibration curve was linear (r=0.9992) in the range of 0.59~59.0 mg/L, the average recovery was 100.6% (RSD=1.56%). For baicalin, the calibration curve was linear (r=0.9999) in the range of 1.128~112.8 mg/L, the average recovery was 101.5% (RSD=1.68%). For imperatorin, the calibration curve was linear (r=0.999 9) in the range of 0.135~13.5 mg/L, the average recovery was 100.2% (RSD=3.03%). CONCLUSION The method was sensitive, accurate and can be used in the quality control of compound Hu-huang Spray. |
